SUMMARY:Neural Network Based Variational Inference for Cutting Feedback During Uncertainty Propagation
DESCRIPTION:[vc_row][vc_column][vc_column_text css=””]BDHS Seminar presented by Abhirup Datta\nDepartment of Biostatistics\nJohns Hopkins Bloomberg School of Public Health \nIn many scientific applications\, estimates from an earlier (upstream) analysis are used as inputs for subsequent (downstream) analysis\, and uncertainty in these estimates (often available as posterior samples) needs to be propagated into the downstream analysis without reverse feedback. Cutting feedback methods\, also termed as cut-Bayes\, achieve this by constructing a cut posterior distribution that restricts downstream to upstream information flow. As sampling from the cut posterior is generally computationally challenging\, recently variational inference (VI) methods have been developed to approximate the cut posterior. However\, they need two variational approximations\, cannot directly use samples from the upstream analysis\, and often rely on parametric variational families whose approximation quality in finite samples can be poor. We propose\, NeVI-Cut\, a provably accurate neural network-based variational inference method for cutting feedback. We directly utilize posterior samples available from the first analysis\, reducing approximation errors due to a variational approximation for these parameters and circumventing the need to access the data and model used for the first analysis. Conditioning on each MCMC sample\, we then use normalizing flows (neural network based generative models) to specify the conditional variational family for the downstream parameters. We estimate the conditional cut posterior by optimizing the Monte Carlo average of the Kullback-Leibler loss over all the MCMC samples. Leveraging the universal expressiveness of neural networks\, we provide theoretical guarantees on the ability of NeVI-Cut to approximate any cut posterior. In the process\, we provide a general result on uniform approximation of conditional distributions by flows. A triply stochastic scalable algorithm is devised to implement the method. Simulation studies and two real-world analyses illustrate how NeVI-Cut achieves significant computational and performance gains over traditional cutting feedback methods. \nA seminar tea will be held at 2:45 p.m. in University Office Plaza\, Room 240. SUMMARY:Reproductive Justice Denied: The Structural Violence of Recent Federal Health Care Cuts
DESCRIPTION:[vc_row][vc_column][vc_column_text css=””] \n\nThis presentation examines recent health care cuts through the lens of reproductive justice\, arguing that they build on a history of structural violence within the U.S. health policy landscape. Rather than reduce spending through universal health coverage reform\, recent legislation attacks key pillars of our healthcare system: Medicaid\, the Affordable Care Act\, and reproductive health care providers. This structural violence will widen long standing health inequities affecting low-income and underserved populations\, and particularly immigrants\, Black\, Indigenous\, and People of Color\, and disabled people. Further\, it will raise barriers to reproductive justice: the right to have children\, not have children\, and raise children in safe\, sustainable\, and healthy communities. The injustices to come are not incidental; they are legislated. This presentation offers the Medicaid and Reproductive Justice Collaborative’s “Reproductive Justice Principles for Federal Universal Health Care Reform” as a policy framework for assessing and advancing federal health care reform that centers inclusion\, representation\, and nondiscrimination and ensures affordable and comprehensive health coverage\, including sexual and reproductive health coverage\, for all.\n\n\n[/vc_column_text][/vc_column][/vc_row]

DESCRIPTION:[vc_row][vc_column][vc_column_text css=””] \nObject-on-Object Regression\nPresented by Yi Zhao\nDepartment of Biostatistics & Health Data Science\nIndiana University School of Medicine \nIn this presentation\, two approaches of object-on-object regression will be introduced. The first is to perform a Covariance-on-Covariance Regression (CoCR)\, which transforms covariance matrix objects to Euclidean space for regression; and the second is to perform a Density-on-Density Regression (DoDR)\, which directly defines regression in the correspondingly constructed Riemannian manifold. \nThe CoCR model assumes that there exists (at least) a pair of linear projections on outcome covariance matrices and predictor covariance matrices such that a log-linear model links the variances in the projection spaces\, as well as additional covariates of interest. An ordinary least square type of estimator\, which relaxes the distributional assumption\, is proposed to simultaneously identify the projections and estimate model coefficients. Under regularity conditions\, the proposed estimator is asymptotically consistent. This type of model is motivated by utilizing functional connectivity within the resting-state network to predict brain connectivity within a corresponding task-state network. Applying to data collected in the Human Connectome Project Aging study\, three networks\, corresponding to a global signal network\, a task-related network\, and a task-unrelated network\, are identified. \nThe DoDR model is introduced to elucidate the association between densities via a warping function. The proposed model has the advantage of a being straightforward demonstration of how one density transforms into another. Using the Riemannian representation of density functions\, which is the square-root function (or half density)\, the model is defined in the correspondingly constructed Riemannian manifold. To estimate the warping function\, it is proposed to minimize the average Hellinger distance\, which is equivalent to minimizing the average Fisher-Rao distance between densities. An optimization algorithm is introduced by estimating the smooth monotone transformation of the warping function. Asymptotic properties of the proposed estimator are discussed. Simulation studies demonstrate the superior performance of the proposed approach over competing approaches in predicting outcome density functions. Applying to a proteomic-imaging study from the Alzheimer’s Disease Neuroimaging Initiative\, the proposed approach illustrates the connection between the distribution of protein abundance in the cerebrospinal fluid and the distribution of brain regional volume. Discrepancies among cognitive normal subjects\, patients with mild cognitive impairment\, and Alzheimer’s disease (AD) are identified\, and the findings are in line with existing knowledge about AD. \nA seminar tea will be held at 2:45 p.m. in University Office Plaza\, Room 240. SUMMARY:Modeling Opioid Use Disorder and the Impact of Interventions in the US – a Geospatially Detailed Agent-based Model
DESCRIPTION:[vc_row][vc_column][vc_column_text css=””]Under funding from the Centers for Disease Control and Prevention\, we have constructed a complex model of opioid use disorder in the Framework for Reconstructing Epidemic Dynamics (FRED) an agent-based modeling platform developed by the Public Health Dynamics Laboratory at the University of Pittsburgh School of Public Health.  The model combines data on individual characteristics (age\, gender\, race\, metal health history\, opioid use history) with characteristics from the geographic location in which the individual resides (density of OUD disorders\, amount of Fentanyl\, availability of naloxone and buprenorphine) to estimate the likelihood that an individual progresses through different states of opioid use disorder.  Using data from the literature\, we estimate the impact of various interventions on the number of people with OUD and the number of OUD-related deaths. We are developing county-specific front-end to estimates the impact of combinations of interventions. \n\n[/vc_column_text][/vc_column][/vc_row]

DESCRIPTION:[vc_row][vc_column][vc_column_text css=””]BDHS Seminar presented by Li Chen\nDepartment of Biostatistics\nUniversity of Florida \nUnderstanding gene regulatory networks (GRNs) is essential for deciphering biological processes and disease mechanisms. Single-cell multiome technologies now enable joint profiling of chromatin accessibility and gene expression\, offering a powerful means to infer cell type-specific GRNs. However\, existing methods analyze each cell type independently or aggregate data into pseudo-bulk profiles\, limiting their ability to resolve rare populations and capture cellular heterogeneity. We introduce BayesCNet\, a Bayesian hierarchical model that jointly infers enhancer-gene linkages across all cell types while leveraging their hierarchical relationships for information sharing. Through extensive simulations\, BayesCNet consistently outperforms state-of-the-art methods\, with the largest improvements in rare cell types. When applied to real datasets\, BayesCNet identifies enhancer-gene linkages with higher accuracy validated by promoter-capture Hi-C data\, and reconstructs cell type–specific GRNs that highlight key regulators\, demonstrating its power to resolve gene regulatory programs across diverse cell types. \nA seminar tea will be held at 2:45 p.m. in University Office Plaza\, Room 240.
